Tirzepatide glucose trials at mypeptidesrx
The diabetes comparison and the obesity comparison both favour tirzepatide. Their measurements—and the questions they answer—deserve separate space.
The diabetes comparison: SURPASS-2
HbA1c is the blood marker used to track longer-term glucose control. In SURPASS-2, an open-label trial in 1,879 adults with type 2 diabetes, tirzepatide reduced it by 2.01-2.30 percentage points across the study’s three doses, versus 1.86 points with semaglutide 1 mg, over 40 weeks. Weight reductions were also greater with tirzepatide.
The comparator narrows the conclusion I draw from this experiment. The study tested a specified semaglutide dose for a diabetes question. Stretching its result into a verdict on every semaglutide formulation or every clinical goal seems too broad to me. The direct evidence is already impressive without that extension.
A fall in HbA1c is reported in percentage points. It is a different measurement from percentage body-weight change.

The obesity comparison: SURMOUNT-5
SURMOUNT-5 supplied the more direct answer to the weight-loss question. It enrolled 751 adults with obesity without type 2 diabetes and compared maximum tolerated doses over 72 weeks. Mean body-weight change was -20.2% with tirzepatide and -13.7% with semaglutide (P<0.001).
The placebo-controlled SURMOUNT-1 trial had already shown substantial weight effects in 2,539 adults with obesity or overweight and a weight-related complication, without diabetes. Its study arms produced mean changes of -15.0%, -19.5% and -20.9%, versus -3.1% with placebo, at 72 weeks. Gastrointestinal adverse events occurred mainly during dose escalation.
I regard the direct comparison as more useful for choosing between those two trial treatments than lining up unrelated placebo trials. Even so, a stronger average weight result leaves the separate questions of tolerability, glucose goals and clinical suitability open.
Dual receptor action
Tirzepatide is a single peptide that activates GIP and GLP-1 receptors. These gut-hormone pathways affect glucose-dependent insulin release, gastric emptying and appetite. Its clinical reference chapter describes the mechanism and the original diabetes indication.
The dual action invites investigation. I hesitate when it is used to presume superiority everywhere. The trials establish measured glucose and weight outcomes; they do not fully divide the benefit between the two receptors.
Tirzepatide is FDA-approved for type 2 diabetes, chronic weight management, and moderate-to-severe obstructive sleep apnea in adults with obesity. The approvals occurred in different clinical settings. The reference chapter’s discussion of weight loss as off-label reflects its account of the original diabetes indication; the site’s current approval summary includes weight management.
Pancreatitis and biliary findings
The nine-trial safety meta-analysis pooled 9,871 participants. It found an increased risk of the composite of gallbladder or biliary disease versus controls (RR 1.97, 95% CI 1.14-3.42). Individual components did not each show a significant increase.
For pancreatitis, the estimate was RR 1.46, 95% CI 0.59-3.61, without a statistically significant increase. I hesitate over calling the pancreatitis question closed: the interval allows considerable uncertainty. The gallbladder signal, meanwhile, deserves space beside the favourable efficacy numbers.
The clinical reference also describes the rodent-derived thyroid C-cell-tumour boxed warning and the contraindication for personal or family history of medullary thyroid carcinoma or MEN-2. Gastrointestinal effects were the most common adverse events in SURPASS-2 and SURMOUNT-1.
Accounts of everyday treatment
People describe a quieter appetite, changes in energy, improved sleep and better mood, alongside nausea, bowel changes, sulfur burps and injection-site reactions. These are anecdotal accounts, not clinical evidence; a collection of reports cannot establish an effect’s frequency or its cause.
What interests me in those accounts is the range of experiences hidden by an average weight change. A person may value glucose control, easier movement or sleep differently. The comparison page returns to measured outcomes, with a graphic separating the diabetes and obesity comparisons.
Peptides RX LLC
The tirzepatide studies summarised on this page concern the compound. Their findings do not establish who “Peptides RX LLC” is or what it offers; the cited sources leave those entity questions unanswered.
